Plot: Nathan Drake (Ryan Reynolds) and Victor "Sully" Sullivan (Bryan Cranston) are drinking in a bar in London, England, waiting for someone. Sully asks Drake why he got married if he's afraid of being a husband. Drake struggles to answer the question. A man named Talbot (Richard Armitage) sits down across from them and asks to see Francis Drake's ring. Drake takes it off and sets it on the table. Talbot then sets an envelope of bank notes on the table. Sully starts inspecting the banknotes, and accuses Talbot of trying to buy the ring with counterfit notes. They get up and take the ring back, but Talbot has several body guards swarm the table. Drake grabs a chandelier and swings his way toward the guards, kicking them to the ground. Sully breaks a beer bottle over a guard's head. Drake and Sully race out the back door of the bar and into an alley way where they are quickly tased by Charlie Cutter (Mark Strong). Drake and Sully hit the ground hard. Talbot emerges from the bar alongside Katherine Marlow (Helen Mirren). Marlow takes the ring from Drake and gets into a car with Talbot. Drake gets up to chase after her, but he and Sully are shot by Cutter.

20 Years Earlier. A Young Nathan Drake (Jack Dylan Grazer) is exploring a museum in Cartagena, Colombia, when happens upon the display of Sir Francis Drake's ring. Drake watches a slightly less grizzled Victor Sullivan quickly steal the ring from the display. Drake follows Sully out of the museum, bumping into him in the streets in order to pickpocket the ring from Sully. Drake walks away, but is stopped by Katherine Marlowe and Sully, who works for Marlowe. Marlowe tells her guards to take Drake out, but Sully stops them and allows Drake to get away. That night, Drake is camped out on a rooftop when he is approached by Sully. Sully tells Drake that he has some skill, but needs someone to look after him.

Back in the present, Cutter helps Drake and Sully up off the ground. Drake and Sully rip the spent squibs off of their chests and thank Cutter for his help. A van then pulls up driven by Chloe Frazer (Sofia Boutella). Drake, Sully and Cutter hop into the van. Frazer tells them that she put a tracking device on Marlowe's car and begins following the car's location. They drive until they are 10 feet from the car according to the tracker, but there is a brick wall in their way. Sully comments that they probably have to go on foot from here.

Drake and Chloe scale the wall and reach a window. Sully and Cutter wait outside, saying their too old to climbing up the sides of buildings. Inside the building, Drake and Chloe find a library. Drake starts looking around while Chloe keeps watch. Drake finds a notebook belonging to T.E. Lawrence and a mpa showing Francis Drake's secret voyage to Arabia. Hearing people coming, Drake pockets the items, and he and Chloe escape out the window. Back at their hotel, Drake, Chloe, Sully and Cutter discuss the clues. Drake explains that the map shows Francis Drake's voyage to Arabia where he was commissioned by Queen Elizabeth I to search for the lost city of Ubar, aka Iram of the Pillars, a place that is only mentioned in the Quran. He tells the group that he's heard legend of this expedition, but the map will not lead them to the location alone. They will need to access Crusader crypts in France as well as Syria. The group decides to split up, with Sully and Drake heading to France, while Chloe and Cutter travel to Syria. Once Drake and Sully are done in France they will meet up with the others in Syria.

Drake and Sully head to eastern France and locate the long abandoned chateau in a dense forest. They break into the crypt, finding one half of an amulet. Suddenly they are ambushed by Talbot and a group of mercenaries. Talbot takes the amulet piece and locks Drake and Sully in the chateau. Talbot then has his men set fire to the chateau. Drake and Sully make their way up the stairs of the building, away from the fire on the first floor. From an upper window, they see that they can jump to a nearby tree. Drake and Sully jump over to the tree, which itself catches fire and falls over. Drake and Sully fall along with the flaming tree, hitting the grass with a hard thud. Sully has the wind knocked out of him. Drake grabs him and helps him away from the fire and back through the forest, which has also caught flame. Drake and Sully make it to a small boat along a river. Drake and Sully know they now have to get to Syria as soon as possible since Talbot and his men may be after them next.

In Syria, Nate and Sully meet up with Chloe and Cutter. Cutter tells them that he and Chloe learned in the crypt that Marlowe is the head of a secret order dating back four centuries. Chloe says that the order seeks to gain power by exploiting the fears of their enemy. The group heads back to the crypt at a local citadel where they find the other half of the amulet, which reveals that the next clue they must find is located in Yemen. Sully rolls his eyes, expressing frustration that the treasure is never just in the first or second location, that they always have to go to several places first. The group begins exiting the citadel when Talbot and Marlowe emerge with a team of mercenaries. Talbot corners Cutter and forces him to hand over the amulet piece. Drake, Sully and Chloe climb over a stone wall and yell out to Cutter. Cutter runs to join them, climbs over the wall and jumps down to the other side, but Cutter breaks his leg on the landing. Chloe tells Drake and Sully to get to Yemen, while she helps Cutter escape the area.

While Drake and Sully are on a train to Yemen, Sully asks Drake if he realizes that his wife Elena is in Yemen right now. Drake says he knows and is nervous about seeing her. When they arrive in Yemen, Sully orders Drake to go see Elena, telling him that treasure will wait, but love will not. Drake begrudgingly agrees. Drake meets up with Elena Fisher (Brie Larson) and notices that she is still wearing the wedding ring he got her. Elena asks Drake what quest he's on today, and he tells her that he is looking for the location of a lost city. Elena is not surprised at all. Elena asks if Sully is with him, and Drake says he's down the street. They go to meet up with Sully. Elena gives Drake and Sully a tour of the city. Sully then shows Elena the map and asks Elena if she can help them find the location shown on the map. Elena begrudgingly agrees and the three of them follow the map to some ruins outside of the city. Drake looks around until he sees a familiar symbol. He kicks the symbol, which opens up a tomb. Drake searches inside the tomb and finds evidence that Sir Francis Drake had also found the tomb, but appears to have abandoned his mission at that location.

When Drake comes back above ground he is shot by a dart. He stumbles away from the ruins in a daze. He passes out. When he wakes up he finds himself outside a cafe with Marlowe and Talbot. Marlowe threatens Elena if Drake does not cooperate with them. Talbot looks at Drake's phone and sees a text from Sully giving his location. Talbot leaves to go find Sully and Elena. Drake breaks free and chases Talbot through the city. Drake, however, is knocked out and captured by Rameses (Oded Fehr), a mercenary working for Marlowe. Rameses interrogates Drake for information about Iram of the Pillars, but Drake refuses to cooperate. Rameses then claims that his friend Sully has been captured and leaves the room. Drake breaks free from his ties. He picks the lock of the cell he is being held in. He sneaks through the building, overhearing that Sully is being held on a cruise ship nearby.

Drake finds the cruise ship and swims out to it. He climbs on board. Rameses and his men appear, revealing that Rameses never had Sully. Drake grabs a gun a shoots Rameses, hitting him in the shoulder. Drake then takes a grenade from one of the mercenaries and throws it. The resulting explosion ruptures the ship's hull. The ship begins sinking rapidly. Drake then swims back to shore.

Back in the city, Drake manages to find Elena. She informs him that Sully was captured by Marlowe's men and taken on a convoy into the desert. Together Drake and Elena hatch a plan to catch up with the convoy by stowing away on a cargo plane due to deliver supplies to the convoy. At dawn, Drake and Elena infiltrate the local airstrip. They reach a high wall, which Drake easily scales, but he refuses to help Elena up. He tells her that he doesn't want to risk her getting hurt. Elena says she understands and walks away. Drake continues toward the plane. Elena notices a jeep nearby and gets behind the wheel. Drake sees the plane start to take off and runs after it, but he isn't fast enough. Elena then speeds up to him in the jeep, telling him to get in. She catches up with the plane, allowing Drake to climb through a landing-gear bay door just as the plane lifts off. Drake is discovered by Marlowe's mercenaries onboard, leading to a shootout. The exchange of gunfire causes a fire onboard the plane, leading to big change in air pressure. The plane then begins to tear apart. Drake grabs onto a large supply crate that is falling from the plane. As the crate is falling through the sky, Drake manages to deploy the crate's parachute, safely landing on the desert floor. Drake begins journeying through the desert.

Drake is suffering from heat exhaustion and thirst after wandering in the desert. He is surrounded by Marlowe's men in jeeps. Drake falls to his knees in defeat. Suddenly, a group of men on horseback arrive and fight off Marlowe's men. Salim (Shahab Hosseini) gets off his horse and walks toward Drake, but Drake passes out from exhaustion. Drake wakes up at Salim's encampment. Salim explains that the city of Ubar was doomed thousands of years ago by King Solomon when he imprisoned an evil djinn in a brass vessel and cast it into the city. Salim agrees to help Drake and leads him to the convoy. Drake and Salim destroy the convoy and rescue Sully. Salim then has Drake and Sully follow him into a sandstorm. Drake and Sully lose him in the dust, but then realize that they have arrived at the gates of Ubar.

When they enter Ubar, Drake and Sully find a large water fountain. Drake drinks from it. While he's drinking, Talbot appears and shoots Sully. Sully's lifeless body hits the ground. Drake chases after Talbot, but is attacked by his men who appear to have been possessed by the evil djinn. The mercenaries are engulfed in flames and are able to teleport around the area. Drake starts to realize he's hallucinating and finds Sully alive. Drake realizes that King Solomon must have cast the brass vessel into the water beneath the city, tainting the water with a hallucinogenic property. The city must have gone mad. Drake tells Sully that Sir Francis Drake must have figured out what the vessel was and decided to not bring it back to England knowing what it does to people.

Drake and Sully search the city and locate Marlowe. She has her men using a winch to pull the brass talisman from the water. Drake and Sully sabotage the winch, causing it to explode. The explosion sets off a chain reaction throughout the entire city, causing it to begin to collapse. Drake and Sully begin escaping the city, but encounter Marlowe and Talbot. Suddenly the ground beneath Marlowe collapses, and she falls into a pit of quicksand. Drake tries to save her, but is unable to pull her out. Marlowe pulls Francis Drake's ring off of Drake's hand, taking it down into the quicksand with her.

Drake and Sully are almost out of the city, which is quickly falling into a giant sinkhole. They are then attacked by Talbot, who attempts to kill Drake and Sully. After a long struggle, Drake manages to shoot Talbot. Talbot then falls into the sinkhole. At the city gates, Drake and Sully are met by Salim, who guides them to safety and back to encampment where Elena is waiting for them. Drake runs up to Elena and gives her a hug. He then pulls his wedding ring out of his pocket and puts it back on. Sully asks if there is a plan to get out of the middle of the desert.
