Plot: Treasure hunter, Nathan Drake (Ryan Reynolds), and TV show host, Elena Fisher (Brie Larson) are on a boat off the coast of Panama, having just recovered the coffin of legendary explorer, Sir Francis Drake, Nate's apparent ancestor who was buried at sea over 400 years ago. Elena films Nate as he opens the coffin, only to discover that it is empty except for Sir Francis' diary. Nate radios his mentor and partner, Victor "Sully" Sullivan (Bryan Cranston), asking him to pick them up. When Elena asks what's wrong, Nate says "pirates". Nate arms himself, and gives a gun to Elena just before several pirate boats arrive and begin to climb aboard. After a brief shootout, Sully arrives in his seaplane. Nate and Elena jump overboard and swim to the seaplane, where they are rescued by Sully. As they fly back to the mainland, Nate shows Sully the diary of Sir Francis, theorizing that Sir Francis faked his death and he must have been on to something big.
Nate and Sully are aboard another boat at the pier, while Elena is outside talking to her producer about what just happened to the boat. Nate shows Sully the diary and explains to him that Sir Francis must have hidden the treasure. Sully grows impatient and asks Nate to cut to the chase, so he shows him a page with the words "El Dorado," which is believed to be a fabled lost city of gold. Nate notes that the last page of the diary was torn out, but they both are still eager to go after the treasure. Sully does not want Elena to come. They end up driving away in their boat and leaving her on the dock.
After abandoning Elena at the docks, Nate and Sully begin their search for El Dorado in the jungles of Central America. Following the map in Francis Drake's diary. Sully becomes angered when the treasure isn't where the map showed, and mentions that he needs the treasure to pay off the many debts he has accrued. They find an empty spot where a statue once stood, and realize that El Dorado is not a city of gold, but a golden statue. They continue on through the jungle until they reach a waterfall with a German U-boat hanging over its tip. Before investigating, Nate gives Sully Francis Drake's diary to hold on to while he searches the submarine. He finds a body with pockets full of Spanish gold. Nate finds a map which shows the location of an island that the Spanish took the El Dorado statue to, along with a smaller hand-drawn map that shows the island in detail. As he informs Sully of his discovery over his radio, Sully is suddenly cut off. Nate leaves the boat to find out what is going on.
Nate exits the submarine only to discover Sully is being held at gunpoint by Gabriel Roman (Kevin Costner), Atoq Navarro (Rodrigo Santoro) and his mercenaries. Roman informs Nate that Sully owes him a lot of money. Nate is forced at gunpoint to relinquish the map to Roman and Navarro, although he keeps the second map that he found. Roman feels that Sully needs a reminder and aims his gun at Nate. As Sully tries to discourage him by telling him that Nate has nothing to do with it, Roman shifts his aim to Sully without hesitation and shoots him in the chest.
As Sully falls to the ground, the U-Boat falls down from the waterfall and explodes, catching Roman and Navarro off guard and knocking them over. Nate glances at Sully's body before running back into the jungle. He runs into Elena, who greets him with a punch to the face for leaving her behind. He informs her of Sully's death, before telling her that they have to escape. She apologizes and gives him a pistol. The two fight their way back through the temple, killing several of Navarro's mercenaries.
Nate and Elena travel in Sully's plane to an island in the South Pacific, which is the location marked on the map that Nate discovered inside the U-boat. Elena films their approach for a new episode of her show. Without warning, they are hit by an anti-aircraft fire which destroys one of their engines. Nate tells Elena that they must bail out, and to look for parachutes while he desperately tries to keep control of the plummeting plane. Elena finds two parachutes. In the hurry to get off the plane, Nate accidentally leaves the map behind. He jumps and opens his parachute, only to discover that it has been pierced and has a hole in it. He plummets to the jungle below.
Nate awakens to find himself dangling from a statue, still strapped into his parachute. He cuts himself loose and ventures into the jungle, heading towards the distant column of smoke rising from the wreck of his plane. He eventually finds the front end of the plane in an encampment and is relieved to see that the map is still in one piece. As Nate treks through the island, he stumbles across a large old fortress. He makes his way toward a small ruined tower that he sees her parachute hanging off of. Upon arriving, he finds the parachute harness empty.
Nate finds himself at the base of a large tower. Noticing that the same tower has been drawn on the map that he recovered from the U-boat. Pushing further into the fortress, Nate finds Francis Drake's telescope, and uses it to study a distant harbor, where he sees that the ships that brought Francis Drake to the island lie ruined and almost completely sunk in the water. It dawns on him that Francis Drake may never have made it off the island. He also notices Elena, who is standing near the base of the tower, filming the same sight across the harbor. Eddy Raja (Edi Gathedi) has two of his pirates, one armed with a grenade launcher, sneak up to a ledge behind her and prepare to kill her. Nate opens fire on them, and the one with the grenade launcher turns and fires at him instead. The resulting blast destroys a portion of the tower and knocks Nate unconscious.
He is awoken in a jail cell by Elena, who is throwing stones in at him through the barred window. She informs him that it will just take a tug to pull the bars out, and disappears as Eddy Raja enters the jailhouse, carrying Nate's map. Nate refuses to cooperate with Raja's threats and demands, saying that he'd rather die than help him. While the two have been talking, Elena has placed a hook on the cell window bars, and attached the hook to a jeep using a cable. She uses the jeep to rip the back wall out of the cell, and as Eddy stares in shock, Nate grabs the map from his hands and escapes through the wall. He climbs into the jeep and the two drive into the jungle, pursued by Raja's men.
Nate and Elena escape from the fortress in a jeep, pursued by Eddy Raja's pirates in a jeep chase that takes them across the island. After evading the last of the pirates, Elena almost drives the jeep over the edge of a cliff, and it is left hanging over the edge. As Nate attempts to drive it back up, Eddy arrives and forces them to get out and surrender the map. Nate tries to stall him for a minute, and then reverses the jeep, backing it off the cliff and into the water below. Nate and Elena swim out of their crashed jeep into a nearby city, which has become largely flooded over the years. Elena shows Nate what she has been filming with her video camera, pointing out a building in the harbor that all cargo coming off ships must have went through. She surmises that the El Dorado statue must have went through there too. Nate notices a small motorboat, and decides that it is their best chance of escaping the island.
Nate and Elena move through the Customs House, a large harbor building that all cargo on the island must have passed through. Nate and Elena enter a room containing ship manifests, and stop to study one that has been left open on a table in the center of the room. It details a statue, labeled El Dorado. He tears the El Dorado page out of the log, points it at Elena's camera and says that it's the closest to El Dorado that they're getting.
They continue on towards the boat, and Elena suggests that they separate so that she can film more of the island, while Nate gets the boat and comes back to pick her up. Nate agrees, and moves onward, entering a large domed building. As he enters, a helicopter flies overhead, and he points out that Eddy's pirates don't have helicopters. The door that he entered through slams shut behind him. Nate is ambushed by a large group of Eddy's pirates. After fighting his way through them, he advances to the motorboat that will get him and Elena off the island. Elena shows him video footage of Roman escorting the supposedly dead Victor Sullivan onto a helicopter with Navarro.
Nate and Elena arrive at a monastery. Once there, they face Navarro's mercenaries, who have overrun the area. Fighting their way through the monastery and surrounding jungle, they come across the body of one of Eddy Raja's pirates impaled on a spike trap. Elena wonders why Raja or Navarro's men would set up traps when they are at risk of triggering them. Nate rightly points out that they wouldn't. Elena questions whether the Spanish had made them hundreds of years earlier, then Nate tells her to look more closely at the spikes, as they have been clearly made from the metal of their plane, indicating that they were made very recently. They begin to have bad feelings about the place, and insists they hurry up and find Sullivan so they can get out of there. They shoot their way into a building, and confront Sullivan about his supposed death. He shows them Francis Drake's diary, which he had placed in his shirt's breast pocket, allowing it to stop the bullet from killing him. After assuring Elena that they will not leave her behind again, the three decide to go for the treasure. Nate discovers a passage to a labyrinth of catacombs beneath the monastery. He tells Sullivan and Elena to wait in a hidden room where they will be safe. He carries a walkie-talkie to communicate with them, and Elena warns him to be careful. They close the door back up, and Nate heads down into catacombs. Eventually, Nate reaches a grating beneath a small mausoleum where Gabriel Roman and Atoq Navarro are arguing with Eddy Raja. Raja tells them that his men are being massacred by something else and claims that the island is cursed. Roman dismisses him and tells him that their contract is over, and that Eddy will not receive a share of the treasure. Eddy pulls his gun on Roman, only to be stopped by Navarro.
While traveling through the tunnels, Sullivan is separated from Elena and Nate when a trap door slams down. Nate and Elena arrive into a small circular room at the top of a treasure vault. Expecting to find El Dorado, they instead discover the remains of Francis Drake. Nate is clearly disappointed that Drake never found the treasure, and leaves his ring with the body. Nate and Elena start looking for a way out, as the distant screams of Eddy Raja's pirates begin to echo through the area. Nate boosts Elena up to a broken ladder, where she climbs up to the room's upper level. She tells Nate that there is a rope attached to an old hoist, but discovers that it is stuck as she attempts to swing it out to Nate. As she attempts to move the hoist, Eddy storms into the room, clearly terrified. Nate aims his gun at him, but is interrupted by nearby roaring and the screams of Eddy's dying pirates. Nate asks Eddy what's happening, when suddenly human-like creatures, known as "descendents" begin to pour into the room through holes in the floor, and Nate and Eddy are forced to work together to stay alive. The two open fire on the descendants.
During the firefight, Eddy kills a descendant whose body falls into one of the holes. Proud, he moves to the edge of the hole. Another descendant then reaches out of the hole. Eddy is bitten in the neck and pulled down into the hole. Nate is faced with several more descendants that appear. He continues to fight alone until Elena manages to move the hoist and get the rope out for him to climb up. The two run down a passageway, pursued by the descendants. They eventually pass through a large metal doorway, which they close and seal shut behind them. With the descendants roaring outside, Elena is the first to notice their location; they are in an upper control room of a long abandoned German submarine bunker. Nate runs up to the window and spots a submarine sitting in a dry dock far beneath them. Nate searches the bunker, finding a projector, and play footage of the German expedition. The footage shows the El Dorado statue, along with one of its victims. A man has been chained to it, and is attempting to attack the camera, behaving exactly like the descendants. On the table beside the projector, Nate finds a letter: the last testament of Francis Drake. In the letter, Francis Drake explained that the treasure bears a terrible curse, and that it was he who destroyed the Spanish ships in an attempt to stop it from ever leaving the island. Reading this, Nate realizes that the treasure also affected the Spanish hundreds of years ago. Through centuries of inbreeding, they became the descendants that were currently roaming the island. Nate sees Elena being captured by Roman and Navarro. Roman thanks him for leading them to El Dorado. They then leave, taking Elena prisoner.
Navarro's mercenaries enter the bunker and try to kill Nate, but they soon get caught up in fighting the descendants. Nate fights through both descendants and mercenaries before climbing a ladder that takes him back up to the monastery above. Nate contacts Sully over his walkie-talkie after escaping the bunker. He tells him that Roman and Navarro captured Elena. Sully then tells Nate that he's pinned down in the monastery courtyard. Nate comes to his aid, and knowing that he cannot let the cursed El Dorado leave the island, Nate once again enters the catacombs beneath the church, with Sully close behind.
Nate and Sully move through the deserted catacombs and arrive in an open cavern beneath the graveyard. Roman is inspecting the El Dorado statue, which stands beneath a large hole in the ground above. To his right, Navarro is holding Elena at gunpoint. As Nate and Sully enter, they are ambushed from behind by two of Navarro's mercenaries. As Roman admires the statue's magnificent craftsmanship, Navarro tells him that it's only a shell, and that the real treasure of El Dorado lies inside. Roman opens the "statue," which is now revealed to be a sarcophagus. Inside, he finds human remains. Before he can turn to Navarro for an explanation, he falls victim to El Dorado's "curse," an airborne virus that within seconds causes him to lose all reason and become aggressive like the descendants. As he runs to attack Navarro, Navarro shoots him in the head. Navarro then orders his men to airlift the statue out of the cavern.
As Nate tries to reason with him, Navarro reveals his true motive: money. Having known about the virus all along, he used Roman's resources to get him to the island. Now, with Roman dead, he plans to sell the virus. As he leaves the cavern with Elena, the descendants begin to swarm in. Nate and Sully take the opportunity to overpower two of the mercenaries, taking their weapons in the process. Nate sets off after Navarro, while Sully remains in the cavern providing covering fire for him. Nate manages to grab the net that holds the statue beneath the helicopter as it takes off. As they fly off the island, Nate is spotted by a mercenary on the ground who radios Navarro on the helicopter. Navarro orders one of his men to shoot Drake. As the man fires out through the open door, Elena kicks him out of the helicopter. He continues shooting as he falls, and accidentally hits the pilot. With the pilot dead, the helicopter plummets toward Navarro's boat. Nate drops off of the statue and lands at one end of the boat, while the helicopter crashes at the other, hanging over the edge. With Elena unconscious, Navarro climbs out of the wreck and orders his men to kill Nate.
Nate fights his way across Navarro's ship, arriving where the helicopter has crashed. As Nate runs to help an unconscious Elena. Navarro attacks him, but Nate eventually knocks Navarro to the ground. Nate then runs to help Elena out of the helicopter, and as she climbs out, she notices that Navarro is getting back to his feet, gun in hand. Nate notices that the cable connecting El Dorado to the helicopter is in a loose loop around Navarro's foot. Nate pushes the helicopter overboard, which plummets into the ocean below, pulling Navarro and El Dorado down with it into the ocean.
Nate and Elena then discuss their experience. As the two lean in for a kiss, they are interrupted by Sullivan. He reveals that he has brought with him a boat load of treasure, which Nate and Elena are happy to see. As they sail back to civilization, Elena reminds Nate that because she lost her camera, he still owes her a story.
