Plot: In the dungeon beneath his estate, Abraham Van Helsing (Michael Fassbender) drains blood out of a coffin and injects it into his veins.

Thomas Carnacki (Hugh Laurie), inspector with the Scotland Yard, pays Van Helsing, famed monster hunter, a visit for help with a case. There have been a series of murders that he cannot manage to solve through traditional means as he believes that the supernatural is involved. Multiple witnesses have reported seeing a woman murdered in front of them, by some seemingly invisible force.

One of the witnesses is a local medium, Cecile (Hayley Atwell). Van Helsing goes to visit her in her shop. Suddenly she is dragged towards the chimney. Van Helsing gives chase. He fires his crossbow and the force releases Cecile. The invisible force throws Van Helsing and Cecile through the window before escaping.

Van Helsing goes to see Dr. Jack Griffin (Daniel Craig), a famed scientist known in certain circles for his work in the field of refraction. Griffin tells him that it is possible, based on his research, for someone to become invisible, and that he and his colleague Dr. Arthur Kemp had done some work on one before Kemp recently went missing.

Van Helsing believes Kemp could be involved and has Carnacki let him into Kemp's house. In Kemp's cellar, Van Helsing finds that a deep hole has been dug. Van Helsing is pulled down into the hole by the invisible force, which takes him down into the sewers. Th monster drags him into the water, and they travel through the canal until they fall into a cage trap. The invisible force is rendered unconscious from the fall, leading its invisibility to wear off, revealing Dr. Arthur Kemp (Damian Lewis) to be the invisible murderer.

Van Helsing easily escapes the trap, but finds himself surrounded by cages full of various mutilated corpses comprised of various human and animal body parts sewn together. Van Helsing finds documents on a nearby desk that says that an experiment was injected with a serum of invisibility. From out of the shadows, Dr. Alphonse Moreau (Robert De Niro) appears. They discuss Moreau's experiments, with Moreau revealing that he seeks to continue where Dr. Victor Frankenstein left off before he died and take the science to its natural progression: to create new lifeforms. Moreau even reveals to Van Helsing that he had his assistant, Kemp, dig up Dr. Frankenstein's body, in hopes to bring him back to life, or at least utilize his brain. Van Helsing calls him a murderer and a butcher and tells Moreau he is going to arrest him. Moreau is unfazed and continues sharing his vision of creating chimeras to do his bidding. Kemp wakes up and injects himself with more invisibility serum. He then attacks Van Helsing from behind.

While Kemp and Van Helsing fight, Moreau releases his lone successful chimera, a frog-like beast, to attack Van Helsing before escaping into the sewers. Van Helsing successfully subdues Kemp, but decides to not kill the chimera, instead allowing it to escape into the sewers.

Van Helsing leaves Kemp in the custody of Carnacki before going to see Cecile. He asks her what his future holds, but she is startled by what she sees and refuses to tell him.

Meanwhile, the coffin in Van Helsing's office is broken open by Frankenstein's Creation (Edgar Ramirez), revealing it to be the chained-up, but still-alive Dracula (Oscar Isaac).
